Immunotherapy has emerged as a useful treatment option for various cancers and some other disesases. However, cancer cells may utimately develop resistance to this treatment. It we can find ways to prevent this from happening or to solve it, it will greatly improve the effectiveness of immunotherapy.

In a tumor, new blood vessels are generated, and these blood vessels offer oxgen and nutrients that maintain the survival and growth of the tumor cells. These tumor blood vessels also prevent T cells from reach the tumor cells, which protects the tumor cells from the effects of the immune system and immunotherapies. VEGFA and ANGPT2 are two major proteins that are involved in the growth of tumor blood vessels and inhibiting T cell response.

Now a study "Dual angiopoietin-2 and VEGFA inhibition elicits antitumor immunity that is enhanced by PD-1 checkpoint blockade", which is published in Science Translational Medicine, reveals that an antibody called A2V can simultaneously suppress VEGFA and ANGPT2 and metastasis.

An important aim of this study is to determine optimal ways to combine immunotherapies with other cancer treatments. The study is led by Michele De Palma from Swiss Institute for Experimental Cancer Research (ISREC) in Switzerland.

A2V blocks both VEGFA and ANGPT2. Palma's team conducted experiments in models of breast cancer, pancreatic cancer, and melanoma, and found that the antibody showed more significant therapeutic effects than antibodies that inhibit either VEGFA or ANGPT2 alone A2V also suppresses cancer spread, or metastasis.

Palma noted that "A2V not only regressed most of the tumor blood vessels, but also reversed the aberrant features of those that had remained, making them similar to normal blood vessels and more permissive to the arrival of anti-tumoral T cells."

The study also showed that combination of immune system-activating and anti-angiogenic antibodies resulted in better therapeutic outcomes. (Cusabio provides proteins and antibodies including polyclonal antibody.)
